Handbook of proteomic methods /
作者: edited by P. Michael Conn.
简介:Summary: Publisher Summary 1 Provides material lists and step-by-step instructions for conducting proteomic experiments in the laboratory, particularly the identification of proteins by mass spectroscopy. The 34 chapters describe general techniques, post-translation modifications, variant separations, protein isoforms, methods for studying specific systems, and tools for data analysis. Topics include multicolor detection on Western blots, a strategy for characterizing antibody/antigen interactions using ProteinChip arrays, stable isotope labeling with amino acids, predicting glycan composition from experimental mass, quantitative characterization of proteomics maps by matrix invaritants, and patchwork peptide sequencing. Annotation (c) Book News, Inc., Portland, OR (booknews.com) Publisher Summary 2 A compendium of thirty-four powerful techniques for identifying and analyzing the diversity of proteins expressed in cells. Thee readily reproducible proteomic methods range from general to specific techniques, and include methods for data analysis, posttranslational modification, and its variants and isoforms. Additional methods demonstrate the application of proteomics to the discovery of serological tumor markers, to identifying the determinants of sensitivity to antitumor drugs, and to specialized fields, such as endocrinology, plant biology, nephrology, and urology.

简介:The critically acclaimed laboratory standard for more than forty years, Methods in Enzymology is one of the most highly respected publications in the field of biochemistry. Since 1955, each volume has been eagerly awaited, frequently consulted, and praised by researchers and reviewers alike. Now with more than 300 volumes (all of them still in print), the series contains much material still relevant today--truly an essential publication for researchers in all fields of life sciences. Major Sections Include: * Basic Principles, Specialized Uses, and Genetic Applications* LCM and its application in genomics and proteomics * Fluorescence in situ hybridization of LCM isolated nuclei from paraffin sections* Noncontact laser catapulting for the functional genomics and proteomics * Use of LCM for clonal analysis, in carcinoma analysis, to assess development, in complex tissue, in pathology, gene discovery, and more.

简介:This volume supplements Volumes 63, 64, 87, and 249 of Methods in Enzymology. These volumes provide a basic source for the quantitative interpretation of enzyme rate data and the analysis of enzyme catalysis. Among the major topics covered are Engergetic Coupling in Enzymatic Reactions, Intermediates and Complexes in Catalysis, Detection and Properties of Low Barrier Hydrogen Bonds, Transition State Determination, and Inhibitors. 简介:Summary: Publisher Summary 1 One of the most important recent discoveries in science is the green fluorescent protein, isolated from a bioluminescent jellyfish. The gene that codes for this protein is active in any type of cell, from microbes to humans, producing a "day-glo" dye visible as a bright green light in the microscope. The power of the gene is that by using recombinant DNA technology. GFP can be spliced to any protein, essentially painting that protein green. When such a spliced protein is inside a cell. Further, time-lapse microscopy can be used to examine how that protein and the cellular components that it is part of move within the cell during the cell's life cycle. This Methods in Enzymologyvolume deals with the utility of green fluorescent protein (GFP). The OVID database (including MEDLINE, Current Contents, and other sources) lists nine references to GFP for the ten-year period 1985-1994. In contrast, in less than four years thereafter, over 500 references are listed, a testament to the rapid growth of interest in this probe. This volume documents many diverse uses for this interesting molecule in disciplines that broadly span biology. The methods presented include shortcuts and conveniences not included in previously published sources. The techniques are described in a context that allows comparisons to other related methodologies-such comparisons are valuable to readers who must adapt existing procedures to new systems. Also, so far as possible, methodologies have been presented in a manner that stresses their general applicability and potential limitations. The volume provides a substantial and current overview of the extant methodology in the field and a view of its rapid development.
